The Principal Scientist, Lentivirus Upstream Process Development, will lead the design, optimization, and scaling of upstream production processes for lentiviral vectors (LVV) at Legend Biotech's Somerset, NJ facility. This role is integral to delivering high-quality vector products that support LVV generation and advanced cell therapy programs. The successful candidate will drive scientific excellence in upstream processing, ensuring robust, scalable, and GMP-compliant manufacturing processes.

Key responsibilities include developing and optimizing upstream processes such as seed train expansion, suspension mammalian cell culture (HEK293/HEK293T), plasmid transient transfection, and bioprocessing in bioreactors. The Principal Scientist will establish and validate bioreactor scale-up and scale-down models, translate bench-scale processes to GMP manufacturing, and provide troubleshooting support during clinical production runs. Additionally, the role involves leading study protocols to define operating parameters and performance limits, and collaborating with cross-functional teams to ensure integrated process outcomes.

Candidates should possess an advanced degree (PhD preferred, or MS) in Chemical Engineering, Biomedical Engineering, Virology, Molecular Biology, or a related field, with 8+ years (PhD) or 10+ years (MS) of experience in process development or manufacturing within a biotech or pharmaceutical setting. Deep technical expertise in mammalian cell culture, seed train expansion, feeding strategy, and transient transfection optimization for viral vectors is essential. Proven experience in developing and implementing bioreactor scale-up and scale-down models, a strong understanding of GMP manufacturing requirements, and excellent leadership and communication skills are also required.
