Upstream Scientist
Legend Biotech is seeking an Upstream Scientist to join the Research & Development team in Philadelphia, PA. This role focuses on developing and optimizing upstream processes for lentiviral vector production, supporting in-vivo CAR-T programs. The successful candidate will work closely with cross-functional teams to ensure efficient technology transfer and process alignment.
Key responsibilities include defining and optimizing seed train strategies from shake flask through bioreactors, scaling up to 40-liter systems with reproducible performance. The scientist will execute and optimize lentiviral vector production workflows, including transient transfection and stable cell line optimization. Additional duties involve media optimization, feeding strategy development, and process control adjustments to improve yield, quality, scalability, and robustness. The role also requires developing and executing studies to support scalability and readiness, collaborating with internal teams for technology transfer, and preparing technical reports and study summaries.
Candidates should possess a BS degree with 5+ years, an MS degree with 3+ years, or a PhD with 1 year of industry experience in Bioengineering, Virology, Cell Biology, Chemical Engineering, or a related field. Demonstrated hands-on experience in lentiviral vector production and characterization is required, along with strong experience in mammalian suspension cell culture and upstream bioprocessing. Experience scaling processes to bioreactor systems up to 50 liters is preferred.
